Sterile Technique
The habits, tools, and workflow that let a small mushroom lab work cleanly enough to grow the same strain for years.

Why Sterility Matters
Every mushroom substrate is also an ideal food for bacteria, molds, and yeasts — most of which grow faster than mycelium. A single Trichoderma spore in a sterilized grain jar can outcompete a healthy inoculation within days. At the lab bench, sterility isn't about paranoia; it's about giving your chosen strain a head start that competitors can't erase. The whole discipline of cultivation upstream of fruiting blocks is really the discipline of exclusion.
Contamination Sources
Air: the largest reservoir — every cubic foot of indoor air carries thousands of spores and bacterial cells. Your body: skin, breath, clothing, hair. Tools: unsterilized needles, blades, and spoons. Water: tap water carries bacteria and chlorine-resistant molds. Substrate: incomplete sterilization leaves surviving spores that outcompete your inoculant. Prior work: cross-contamination from an infected jar on the same shelf. Knowing the sources is half the battle.
Clean Workspace Principles
Small, wipeable, uncluttered surface. Airflow moving from clean toward less clean, never the reverse. Isopropyl alcohol (70 percent) is your friend — wipe hands, tools, jar lids, work surface, and outside of bags immediately before opening. Flame-sterilize scalpels and needles until glowing between transfers. Work with slow, deliberate movements; sudden motion pushes air currents that carry spores. And do all inoculation work in a still-air box or in front of a HEPA flow hood — ambient air is the enemy.
Tools and Equipment
Essentials: pressure cooker or autoclave (15 psi for grain and bulk sterilization), still-air box or laminar flow hood, syringes, scalpel, alcohol lamp or torch, 70 percent isopropyl, nitrile gloves, filter-patch bags, quality agar and grain. Upgrades that pay off: a real laminar flow hood, a jar sterilization retort, a dedicated clean room with positive pressure. Every tool exists to close one specific gap in the contamination cycle.
Professional Practices
Batch discipline: sterilize, cool, inoculate, incubate, and inspect in fixed windows so problems are traceable. Isolation: keep spawn work physically separate from fruiting rooms, where wild spores are abundant. Documentation: label everything with strain, date, batch, and inoculator. Regular contamination audits — track your loss rate and investigate spikes. Culture rotation and re-cloning to prevent strain aging. Sterile technique is really quality assurance discipline applied to a biological process.
